    سابقه و هدف

    باکتری های پروبیوتیک نقش بسیار مهمی در بهبود فلور طبیعی روده داشته و مانع رشد باکتری های مضر در دستگاه گوارش شده و ازنظر اهداف دارویی و درمانی دارای اهمیت می باشند. هدف از این مطالعه بهینه سازی تولید زیست توده باکتری لاکتوباسیلوس رامنوسوس GG ATCC53103) Lactobacillus rhamnosus  (با استفاده از روش طراحی آزمون است.

    مواد و روش ها

    در این تحقیق از باکتری پروبیوتیک لاکتوباسیلوس رامنوسوس استفاده شد. به منظور بهینه سازی از روش طراحی پلاکت- برمن استفاده گردید. تمام کشت های محیط پایه و بهینه شده در فرمانتور 1300 لیتری شرکت پارس پاد انجام شد.

    یافته ها

    نتایج نشان داد که منابعملاس چغندرقند، گلوکز و کازیین بیشترین اثر را در تولید زیست توده لاکتوباسیلوس رامنوسوس دارند. گلوکز با کازیین و ملاس چغندرقند اثر هم افزایی داشته و موجب افزایش تولید زیست توده می شوند. پس از بهینه سازی، محیط کشت دارای مقادیر ترکیبات زیر به ازای گرم در لیتر: گلوکز 50/112، ملاس چغندرقند 25/56، کازیین 75/18، عصاره مخمر 75/18، K2HPO4 13/13، تویین 80 88/1، 7H2O.MgSO43750/0، MnSO4. 4H2O 0750/0، CaCl2. 2H2O 1875/0 و سایمتیکن 25/1 به منظور تولید بهترین زیست توده توسط لاکتوباسیلوس رامنوسوس مشخص گردید. میزان زیست توده بیشتر از دو برابر نسبت به شرایط محیط کشت پایه افزایش پیدا کرد.

    نتیجه گیری

    با توجه به تولید زیست توده باکتری لاکتوباسیلوس رامنوسوس در سطح نیمه صنعتی درفرمانتور 1300 لیتری ، امکان تولید صنعتی زیست توده لاکتوباسیلوس رامنوسوس وجود خواهد داشت.همچنین کشت این باکتری به صورت صنعتی در شرایط کشت ناپیوسته تغذیه شونده و متداوم به عنوان یک پروبیوتیک تجاری پیشنهاد می شود.

    سابقه و هدف

    بررسی ساختار جمعیت باکتریایی زیست بوم های پرشور و شناسایی گونه های جدید هالوفیل می تواند از نظر زیست فناوری و اکولوژی حایز اهمیت باشد. این تحقیق با هدف بررسی ساختار جمعیت باکتریایی رسوبات تالاب نمکی جنوب تپه های حلقه دره انجام شد.

    مواد و روش ها

    این پژوهش به صورت مقطعی با نمونه برداری از تالاب نمکی جنوب حلقه دره در خرداد 97 انجام شد. جداسازی باکتری های هتروتروف با استفاده از محیط کشت R2A آگار انجام شد. پس از تفکیک جدایه ها بر اساس خصوصیات مورفولوژیک و بیوشیمیایی، تعیین هویت و ارتباطات فیلوژنتیک جدایه های منتخب با توالی یابی ژن 16S rRNA و بر اساس اطلاعات موجود در بانک ژنی NCBI و توسط نرم افزار های بیوانفورماتیک انجام شد. همچنین از توالی یابی نسل جدید ایلومینا به عنوان روش غیر وابسته به کشت به منظور بررسی تنوع باکتریایی استفاده شد.

    یافته ها

    جدایه ها شامل 13 گونه در 8 جنس باسیلوس (25/31)، هالوموناس(25%)، گراسیلی‏باسیلوس (50/12%)، ویرجی باسیلوس (25/6%)، استرپتومایسس (25/6%)، نیتراتی‏رداکتر(25/6%)، استافیلوکوکوس(25/6%)و پلانوکوکوس (25/6) بودند. همچنین نتایج ایلومینا حاکی از غالبیت گونه های آنورینی‏باسیلوس میگولانس و پانی‏باسیلوس پلی‏میکسا بود.

    نتیجه گیری

    نتایج نشان داد که جمعیت میکروبی تالاب مورد مطالعه با سایر تالاب های پرشور گزارش شده در سایر نقاط دنیا مشابه است و بیشتر جدایه ها مربوط به گونه های هالوتولرانت و هالوفیل بودند. حضور گونه های متنوع می تواند نشان دهنده  وجود گروه های جدید تاکسونومیک و غنای بالای ژنی در این اکوسیستم پرشور باشد که می تواند در پژوهش های تکمیلی مورد بهره برداری قرار گیرد.

    سابقه و هدف

    منابع بیولوژیک رنگدانه ها به دلیل تفاوت آن ها در خواص و شرایط تولید در مقایسه با روش های تولید شیمیایی، مورد توجه پژوهشگران این رشته ها قراراست. هدف از این مطالعه جداسازی کارتنوییدهای تولیدی از گونه های میکروکوکوس و رودوتورولا از منابع خاکی، بهینه سازی شرایط کشت برای تولید زیست توده  و شناسایی رنگدانه کارتنوییدی جدا شده بوده است.

    مواد و روش ها

    این پژوهش به صورت مقطعی توصیفی بر روی 104 نمونه ی میکروکوس و رودوتورولا (جدا شده از خاک مناطق کویری استان کرمان) انجام گرفت. سویه های میکروکوکوس لوتیوس و رودوتورولا موسیلاجینوسا مولد کارتنویید، توسط آنالیز 16S rRNA شناسایی شدند. شرایط بهینه برای تولید بیومس و کارتنویید تعیین گردید. رنگدانه های جدا شده به کمک اسپکتروفتومتری و FT-IR شناسایی گردیدند.

    یافته ها

    شرایط بهینه ی رشد و تولید رنگدانه از لحاظ دما برای هر دو تولید کننده ی رنگدانه دمای 25 درجه ی سلسیوس بوده است. pH برای باکتری ها 7 و برای مخمرها 5/6 شرایط بهینه را ایجاد نمود. یک درصد کربن در محیط کشت برای هر دو (باکتری و مخمر) شرایط بهینه بود و درصد نیتروژن برای باکتری میکروکوکوس لوتیوس دو درصد و برای مخمر رودوتورولا موسیلاجینوسا یک درصد بوده است.

    نتیجه گیری

    داده های به دست آمده در مطالعه حاضر نشان دادکه میکروارگانیسم های شناسایی شده در این مطالعه به عنوان منبع بالقوه بومی خاک ایران در تولید کارتنویید تجاری به حساب می آیند.

    سابقه و هدف
    هیدروکربن های آروماتیک چند حلقه ای، یکی از مهمترین آلاینده های زیست محیطی هستند. پاکسازی زیستی با استفاده از میکروارگانیسم ها، روشی مقرون به صرفه و ایمن جهت حذف و یا تبدیل این آلاینده ها به ترکیباتی با سمیت کمتر است. این مطالعه با هدف جداسازی و معرفی میکروارگانیسم های تجزیه کننده فلورن از سواحل جنوبی دریای خزر انجام گردید.
    مواد و روش ها
    غنی سازی و جداسازی مخلوط میکروبی، در محیط کشت پایه نمکی حاوی فلورن انجام شد. تجزیه کیفی فلورن در محیط پایه نمکی جامد بررسی شد. میزان تجزیه فلورن توسط مخلوط میکروبی نیز با استفاده از کروماتوگرافی گازی در محیط پایه نمکی مایع تعیین شد. شناسایی مولکولی جدایه های باکتریایی و قارچی به ترتیب با تعیین توالی 16S rRNA  و ناحیه محافظت شده ITS انجام گردید.
    یافته ها
    مخلوط میکروبی شامل سویه های باکتریایی (متعلق به جنس های سودوموناس، آکروموباکتر، کریزیوباکتریوم، میکروباکتریوم و رودوکوکوس) و سویه ی قارچی (متعلق به جنس فوزاریوم) غنی سازی و جداسازی شد. آنالیزکروماتوگرافی نشان داد که مخلوط میکروبی قادر است در دمای 30 درجه سلسیوس، اسیدیته 7 و طی 7 روز گرماگذاری، 87٪ از فلورن با غلظت 200 میلی گرم در لیتر را در محیط پایه نمکی تجزیه کند.
    نتیجه گیری
    با توجه به نتایج، مخلوط میکروبی در شرایط یاد شده بخش گسترده ای از فلورن را از محیط پایه نمکی حذف می کند و در شرایط مشابه، ممکن است بتواند از طریق پاکسازی زیستی بخش چشمگیری از فلورن را از منطقه آلوده حذف کند.

    سابقه و هدف

    حضور فلزات سنگین سمی و سرطان زا همچون کروم در فاضلاب صنعتی، یک آلودگی مهم برای خاک های کشاورزی و منابع طبیعی آب محسوب می شود. هدف از این مطالعه ارزیابی حذف کروم با یک جدایه باکتریایی از خاک آلوده در مقایسه با نانوذرات سیلیسی مزوپوری می باشد.

    مواد و روش ها

    در این پژوهش به منظورجداسازی باکتری تجزیه کننده کروم، نمونه برداری از خاک های مناطق مختلف نفتی ساحل کیاشهر صورت گرفت. شناسایی مقدماتی سویه های جداشده بر اساس آزمون های بیوشمیایی و سپس شناسایی مولکولی باکتری با توالی یابی 16S rRNA انجام شد. حذف کروم در شرایط آزمایشگاه با سویه باکتریایی و نانوذرات بطور جداگانه با اسپکترومتر جذب اتمی (AAS) بررسی شد.

    یافته ها

    باکتریتجزیه کننده کروم براساس تجزیه و تحلیل  16S rRNA و همولوژی 99% ، سودوموناس پلکوگلوسی سیدا شناسایی شد. حذف کروم توسط نانو ذرات سیلیسی مزوپور در غلظت 300 میکروگرم در میلی لیتر بیشتر از سویه جدا شده باکتری بوده و میزان حذف را تا 75 درصد نشان داد. اما با افزایش غلظت کروم در محیط به میزان  بیش از  600 میکروگرم در میلی لیتر ، توانایی حذف کروم توسط باکتری نسبت به نانو ذرات سیلیسی مزوپور، به میزان 80 درصد به 72 درصد  عملکرد بهتری نشان داد.

    نتیجه گیری

    نتایج نشان داد که جدایهسودوموناس پلکوگلوسی سیدا و نانوذرات می توانند برای حذف کروم از خاک ها و آب های آلوده استفاده شوند. همچنین به منظور به منظور افزایش کارایی حذف کروم، استفاده هم زمان از باکتری حذف کننده کروم و نانو جاذب استفاده پیشنهاد می شود.

    سابقه و هدف 

    نانوذرات سیلیکا (MCM41) از دسته مزوپورهای سیلیکا (MSNs) دارای منافذ بزرگ و چگالی نسبتا پایین هستند. هدف ازاین پژوهش بارگذاری داروی اکونازول برروی نانوذرات MCM41-NH2 و MCM41 و بررسی رهایش اکونازول در پوست انسان (Ex-Vivo) و اثر ضدقارچی آن بود.

    مواد و روش ها

    در ابتدا MCM41  تهیه شد، سپس با آمین ستیل تری متیل آمونیوم بروماید، نانوذرات MCM41-NH2ساخته شد و داروی اکونازول روی آن بارگذاری گردید. مورفولوژی نانوذرات با دستگاه SEM و بارگذاری دارو با دستگاه FT-IR تعیین و برای اندازه گیری رهایش از دستگاه فرنز سل وUV-Vis استفاده شد. اثر ضد قارچ ECO/MSNs با روش های چاهک پلیت برای قارچ آسپرژیلوس فیومگتس و با استفاده ازروش پورپلیت برروی کاندیدا آلبیکنس بررسی شد. در بررسی رهایش پوستی[p1] [l2]  کرم با نسبت 1 به 1 دارو/ نانوذرات تهیه و از دستگاه فرنز سل استفاده شد.

    یافته ها

    نانوذرات سیلیکا در ابعاد حدود 300 نانومتر شد و رها سازی دارو در پوست نشان داد، طی 8 ساعت اول رهایش80% و پس ازآن تا 24 ساعت رهایش دارو به صورت پیوسته ادامه دارد. بررسی اثر ضد قارچ به روش چاهک پلیت نشان دادECO/MCM41 دارای اثر هاله عدم رشد بزرگ تر و همچنین حداقل غلظت مهاری mg/ml(MIC)75 می باشد. اثرات ضدقارچی پس از 72 ساعت تایید گردید. همچنین حداقل غلظت مهاری  ECO/MCM41 در متانول بیشتر بود.

    نتیجه گیری

     به دلیل اثرات ضد قارچی نانوذرات سیلیسی حاوی اکونازول، استفاده از آن به عنوان داروی مناسب در کرم  پیشنهاد می گردد.

    باکتریوسین های تولید شده توسط باکتری های اسید لاکتیک مانند نیسین فعالیت ضد میکروبی را در غلظت های بسیار کم و بدون عوارض جانبی نشان می دهند. هدف از پژوهش حاضر، بهینه سازی مقدار نیسین در گوشت گاو تخمیر شده برای دست یابی به اسیدیته و جامعه میکروبی بهینه و همچنین کاهش غلظت نیتریت در محصول نهایی بود.در این تحقیق تجربی آزمایشگاهی، ابتدا گوشت گاو تخمیر شده با استفاده از لاکتوباسیلوس پلانتاروم و لاکتوباسیوس ساکیی تهیه شد. سپس بهینه سازی مقادیر نیسین و نیتریت سدیم بر اساس کاهش اسیدیته و تعداد میکروب های عامل فساد در محصول تخمیری با استفاده از روش سطح پاسخ انجام شد. از بین تیمارها، 3 تیمار شامل غلظت های به ترتیب 80 و 100 ppm نیتریت سدیم و نیسین، غلظت های به ترتیب 28/128 و 140 ppm نیتریت سدیم و نیسین و غلظت های به ترتیب 180 و 196  ppm نیتریت سدیم و نیسین بهترین پاسخ را بر اساس کاهش اسیدیته و بار میکروبی مولد فساد نشان دادند. مقدار نیتریت باقی مانده در این سه تیمار به 7/5 تا 47/8 ppm کاهش یافت که کمتر از سطح محدودیت بود. کلاستریدیوم  در هر سه محصول گوشتی پس از نگهداری یک ماهه در 4 درجه سلسیوس مشاهده نشد. افزودن نیسین به گوشت گاو تخمیر شده به طور معنی داری غلظت نیتریت را کاهش داد. بنابراین، استفاده از نیسین به عنوان نگهدارنده در صنایع سوسیس گوشت گاو در ایران توصیه می شود.

    ویروس پالامپور پیچیدگی برگ گوجه فرنگی از بیماری های مهم ویروسی است که از برخی از مناطق کشور گزارش شده است. در این تحقیق تنوع ژنتیکی جدایه های این ویروس از منطقه سیستان و سفیدبالک ناقل آن بیمیزیا تاباسی با استفاده از تعیین ترادف بخشی از ژنوم آن ها تجزیه و تحلیل شد. برگ بوته های دارای علایم این ویروس و حشرات بالغ بیمیزیا تاباسی از شهرستان های منطقه سیستان در فصل پاییز 1398 جمع آوری شدند. ردیابی ویروس با استفاده از آغازگرهای اختصاصی ژن پروتئین پوششی در سه محصول خربزه، بادمجان و فلفل انجام شد. ترادف های نوکلیوتیدی ژن پروتین پوششی سه جدایه این ویروس از منطقه سیستان که از خربزه، بادمجان و فلفل جدا شده بودند یکسانی نوکلیوتیدی 99-92 درصد را با سایر جدایه های ویرس که قبلا گزارش شده بودند نشان دادند. درخت تبارزایی ترسیم شده براساس ترادف ژن پروتئین پوششی نشان داد که جدایه های منطقه سیستان با جدایه های ایرانی گروه بندی می شوند. بادمجان و فلفل میزبان های جدیدی برای ویروس در ایران بودند. تجزیه و تحلیل تبارزایی بر اساس ترادف نوکلیوتیدی ژن زیر واحد شماره 1 سیتوکروم اکسیداز میتوکندریایی نشان داد که نمونه های بیمیزیا تاباسی جمع آوری شده از منطقه سیستان به همراه سایر نمونه های بیمیزیا تاباسی ایرانی در گروه بیوتیپB قرار می گیرند. نمونه های سفید بالک شهرستان های زابل، زهک و هیرمند به ترتیب دارای یکسانی نوکلیوتیدی 100، 75/97 و 68/99 درصد با برخی از نمونه های سفید بالک استان فارس بودند. در این تحقیق جدایه های ویروس از منطقه سیستان با جدایه های ایرانی گروه بندی شدند و نمونه های بیمیزیا تاباسی جمع آوری شده از منطقه سیستان متعلق به بیوتیپ B بودند.

    Background & objectives

     Probiotics play a very important role in improving the normal intestinal flora and inhibit the growth of harmful bacteria in the gastrointestinal tract and are also important for therapeutic purposes. This study aimed to optimize biomass production by Lactobacillus rhamnosus ATCC53103 (GG) using the experimental design process.

    Materials and Methods

     In this study, the probiotic bacterium Lactobacillus rhamnosus was used. Plackett-Burman design method was used for the optimization. Fermentations in basal and optimized cultures were performed in 1300 liter Parspad Company's fermenters.

    Results

     The results showed that beet molasses, glucose, and casein have the greatest effect on biomass production by Lactobacillus rhamnosus. Glucose with casein and beet molasses have a synergistic effect and increasing the concentration of glucose with increasing the concentration of two other factors increases the production of the biomass. Based on the results obtained, after optimization, the optimal culture medium for biomass production by Lactobacillus rhamnosus has the following compounds g/L-1: glucose 112.50, beet molasses 56.25, casein 18.75, yeast extract 18.75, K2HPO4 13.13, Tween 80 1.88, MgSO4. 7H2O 0.3750, MnSO4. 4H2O 0.0750, CaCl2. 2H2O  0.1875 and Simethicone1.25. The biomass production in optimized conditions was increased more than 2-folds higher than the basal medium.

    Conclusion

     Biomass production by Lactobacillus rhamnosus on a semi-industrial scale was carried out in 1300 liters fermentors. Therefore, the results of this study can be used in the industrial production of Lactobacillus rhamnosus biomass. Also, commercial production under fed-batch and continuous culture conditions is recommended.

    Background & Objectives

    Survey of bacterial community structure in hypersaline ecosystems and identification of novel halophilic species can be very important from biotechnological and ecological aspects. In this study, we survey bacterial community structure in sediments from saline wetland in south of Halghe Dare hills as one of the hypersaline ecosystems in Alborz province.

    Materials & Methods

    This cross-sectional study was performed by sampling from saline wetland in south of Halghe Dare hills in June 2018. Isolation of heterotrophic bacteria was conducted using R2A agar medium. After differentiation of isolates based on morphological and biochemical characteristics, identification and phylogenetic relationships analysis of selected isolates were performed by 16S rRNA gene sequencing and analysis using NCBI databases and bioinformatics softwares. The Illumina next-generation sequencing was also applied to survey bacterial diversity by cultivation-independent method.

    Results

    Isolates included 13 species belonging to 8 genera including Bacillus (31.25%), Halomonas 25%, Gracilibacillus (12.50%), Virgibacillus (6.25%), Streptomyces (6.25%), Nitratireductor (6.25%), staphylococcus (6.25%) and Planococcus (6.25%). Illumina sequencing showed that Aneurinibacillus migulanus and Paenibacillus polymyxa were dominant species insoil sample.

    Conclusion

    The results showed that the microbial population of the studied wetland is similar to the community of the wetlands reported in other parts of the world and dominated by halotolerant and halophilic species. Presence of various bacterial species and some probable novel taxonomic groups in saline wetland in south of Halghe Dare hills presents a new genetic and microbial source for future studies.

    Background & Objectives

    Biological sources of pigments receive major attention nowadays because of the stringent rules and regulations applied to chemically synthesized pigments. The aims of this study were isolating carotenoids producing Micrococcus spp < em>. and Rhodotorula spp. from soil sources, optimizing the culture conditions for biomass and carotenoids production and its identification.

    Materials and Methods

    Carotenoid producing strains, M. luteus and R. mucilaginosa, were isolated from the soil and sediment samples in Kerman Province, Iran; they were identified using 16SrDNA analysis. Optimum conditions for biomass and carotenoids production were determined. FT-IR and spectrophotometry analysis showed high similarity of extracted pigments with carotenoids.

    Results

    The optimum temperature of growth and pigment production was 25º C. for both of bacteria and yeasts. The optimum pH for bacteria was 7 and for yeasts 6.5.  1% of carbon source for both of them was the optimum condition while about nitrogen source, 2% for bacteria and 1% for yeasts were the optimum condition for growth and pigment production.

    Conclusion

    Microorganisms presented in this study can be used as potential sources of commercial carotenoids production in Kerman, Iran.

    Background and Objectives
    Polycyclic aromatic hydrocarbons are one of the most important environmental pollutants. Bioremediation using microorganisms is a cost-effective and safe method for the removal or conversion of these pollutants to less toxic substances. This study aimed to isolate and introduce fluorene-degrading microorganisms from the southern coast of the Caspian Sea.
    Materials and Methods
    The mixed microbial culture enrichment and isolation was done in salt-based culture medium containing fluorene. The qualitative analysis of fluorene degradation in the solid basal salt medium was investigated. The rate of fluorene removal by the isolated mixed microbial culture was also determined using gas chromatography in a liquid salt base medium. The molecular identification of the fungal and bacterial isolates was performed using the sequential analysis of the ITS protected region and 16S rRNA sequencing, respectively.
    Results
    The mixed microbial culture including bacterial isolates (belonging to the genus Pseudomonas, Acromobacter, Chryseobacterium, Microbacterium, and Rhodococcus) and fungal isolate (belonging to the genus Fusarium) was enriched and isolated. Chromatographic analysis showed that the mixed microbial culture was able to degrade 87% of fluorene (200 mg / l ) in a basal salt medium at 30 °C, pH 7 and 7 days of incubation.
    Conclusion
    According to the results, the mixed microbial can remove a large amount of fluorene from the basal salt medium under the mentioned conditions and it is possible that under a similar situation, it can remove a large amount of fluorene from the contaminated area through bioremediation.

    Background & Objectives

    The presence of cytotoxic and carcinogenic heavy metals such as Chromium in industrial wastewater is an important pollution for agricultural soils and natural water sources.  The aim of study was to evaluate chromium removal with a bacterium isolate form contaminated soil compared to mesoporous silica nanoparticles.

    Materials and methods

    In this study, for isolate chromium decomposing bacteria, soil were sampled from different  of contaminated beach of Kiashahr. Preliminary identification of the isolated strains was done based on biochemical tests and then molecular identification of bacteria by 16SrRNA sequencing. Cr removal was evaluated with resistant strain and nanoparticles by Atomic absorption spectrometry (AAS), individually.   

    Results

    Chromium-degrading bacteria were identified based on 16SrRNA analysis and 99% homology of Pseudomonas pelicoglucida. Removal of chromium by mesoporous silica nanoparticles at a concentration of 300 μg / ml was higher than the isolated strain of Pseudomonas pelicoglucida and showed a removal rate of up to 75%. However, by increasing the concentration of chromium by more than 600 micrograms per milliliter, the ability of bacteria to remove chromium from mesoporous silica nanoparticles showed better results by 80% to 72% chromium removal.

    Conclusion

    Results indicated Pseudomonas pelicoglucida and nanoparticles can be used to remove chromium from contaminated soils and waters. It is also recommended to use chromium-removing bacteria and nano-adsorbents to remove chromium at the same time to increase the efficiency of chromium removal.

    Background & Objectives

    Silica Nanoparticles (MCM41) are silica mesopores (MSNs), which have large pores and relatively low density. The purpose of this study was to load Econazole onto silica nanoparticles (MCM41-NH2 and MCM41) And release of econazole in human skin (Ex-Vivo) then the antifungal effect of loaded econazole on silica nanoparticles was investigated.

    Materials & Methods

    In this study, MCM41 was prepared, then MCM41-NH2 nanoparticles were provided by using amine cetyltrimethylammonium bromide and finally, the drug econazole was loaded on it. The morphology of the nanoparticles was determined by SEM and drug loading by FT-IR. Francescell and UV-Vis were used to measure the release. The release cream from the skin containing 1/1 drug and nanoparticles was prepared. Finally, the anti-fungal effect of ECO/MSNs was investigated in three ways. Francescell device was also used to check skin release.

    Results

    Silica nanoparticles were prepared about 300 nanometers Release of the drug into the skin showed that during the first 8 hours 80% release and then up to 24 hours of continuous drug release. Antifungal effect by disk diffusion method showed that ECO/MCM41 had a larger inhibitory effect and also minimal inhibition of fungal growth (MIC) 75 mg/ml. Antifungal tests showed that no fungus grew after 72 hours.the antifungal effect as well as ECO / MCM41 (MIC) was greater in methanol.

    Conclusion

    The antifungal effect of the newest agar method was not suitable for this drug. Therefore, as a suitable drug in the cream, a drug loaded with methanol is recommended.

    Bacteriocins such as nisin produced by lactic acid bacteria exhibit antimicrobial activity at very low concentrations without side effects. The aim of the present study was to optimize nisin content in fermented beef to achieve optimum pH and microbial community as well as reduction of nitrite concentration in the final product. In this experimental laboratory research, fermented beef was prepared using Lactobacillus plantarum and Lactobacillus sakei. Then, optimization of nisin and sodium nitrite concentrations were done by response surface methodology based on the pH decrease and spoilage causing microbial count in fermented product. Among treatments, 3 treatments with respectively 80 and 100 ppm sodium nistrit and nisin, 128.28 and 140 ppm sodium nitrite and nisin and 180 and 196 ppm sodium nistrit and nisin showed the best results based on pH and spoilage causing microbial count reduction. The residual nitrite contents in these three treatments reduced to 5.7 to 8.47 ppm, which were lower than the limited amount. Clostridium was absent in all three beef products after one mount storage at 4 °C. Addition of nisin to fermented beef significantly decreased the nitrite concentration. Therefore, the usage of nisin is recommended as a preservative in beef sausage industries in Iran.

    Background & Objectives

    Tomato leaf curl Palampurvirus (ToLCPMV) is one of the most important plant viruses that has been reported from some regions of Iran. In this study, we analysed the genetic diversity of Sistanian isolates of ToLCPMV and their vector, Bemisia tabaci by partial genome sequencing.

    Materials & Methods

    Leaves of plants expressing symptoms of ToLCPMV infection and B. tabaci adults were collected from cities of the Sistan region during the autumn season of 2019. Specific primers of gene coat protein were used to detect ToLCPMV in three crops (melon, eggplant and pepper).

    Results

    Nucleotide sequences of coat protein gene of three Sistanain isolates of ToLCPMV showed 92-99 % sequence identity with previously characterized ToLCPMV isolates. Phylogenetic analyses based on nucleotide sequences of coat protein gene indicated that in the Sistanian isolates of ToLCPMV grouped with other Iranian isolates. Eggplant and pepper represent new natural hosts of ToLCPMV in Iran. Phylogenetic analysis based on mitochondrial cytochrome oxidase-I gene sequences showed that the collected B. tabaci samples from the Sistan region and other Iranian B. tabaci samples belong to the B biotype. Bemisia tabaci samplesof Zabol, Zahak and Helmand counties showed respectively 100, 97.75 and 99.68% nucleotide sequence identity to some from Fars province samples of B. tabaci.

    Conclusion

    In this study, the Sistanain isolates of ToLCPMV were grouped with Iranian isolates of ToLCPMV and the collected B. tabaci samples from the Sistan region belong to the B biotype.
