    هدف:

     سرطان معده چهارمین سرطان شایع در سراسر جهان است که تقریبا در دو سوم کشورهای در حال توسعه رخ می دهد و از علل اصلی مرگ و میر ناشی از سرطان است. MAPK14 نقش مهمی در تبدیل محرک های خارج سلولی به طیف وسیعی از پاسخ های سلولی دارد. HTR2B سطح 5-HT را افزایش می دهد و در نتیجه 5-HT مشتق از پلاکت باعث رگزایی تومور، رشد تومور و پتانسیل متاستاتیک سلول های سرطانی می شود.

    مواد و روش ها

    آنالیز بیان بر روی 20 نمونه سالم و 20 سرطانی بافت پارافینه معده برای MAPK14 و 20 سالم و 19 سرطانی برای HTR2B با استفاده از qRT-PCR انجام شد. پلی مورفیسم در 38 شاهد و 30 مورد برای rs550352538 و 35 شاهد و 35 مورد برای rs28763973 با استفاده از تکنیک T-ARMS PCR بررسی شد.

    یافته ها

    تفاوت معنی داری در سطح بیان ژن های MAPK14 و HTR2B مشاهده نشد (P>0.05). علاوه بر این، هیچ ارتباط آماری معنی داری بین ژنوتیپ های مختلف در rs28763973 از ژن MAPK14 و rs550352538 از ژن HTR2B با سرطان معده وجود نداشت (P>0.05).

    نتیجه گیری

    تفاوت معنی داری بین افراد بیمار و افراد سالم در سطوح بیان و پلی مورفیسم های ژنی مشاهده نشد، با این حال، نتایج ممکن است با تغییر کردن خزانه ژنی یا اندازه جمعیت به طور معنی داری متفاوت شود.

    گلوکوم اصطلاحی است برای بیان گروهی از بیماری ها که دارای ضعف پیشرونده عصب بینایی هستند و باعث کاهش بینایی و در نهایت نابینایی می شوند. بیماری گلوکوم یکی از رایج ترین و بحث برانگیزترین بیماری های چشمی می باشد. علت اختلال بیناییی در این بیماری از بین رفتن تدریجی سلول های گانگلیونی شبکیه، آکسون های آن ها و از بین رفتن همزمان بینایی است. دوپامین که نوعی انتقال دهنده عصبی است، در چشم نقش تعدیل جریان خون مژگانی و تولید آب را دارد، که بر فشار داخل چشم و گلوکوم تاثیر می گذارد. بر اساس مطالعات قبلی بر روی لیگاندهای رسپتورهای دوپامین این نتیجه حاصل شد که اثر دوپامین بر تولید زلالیه با فعال شدن گیرنده شماره یک دوپامین (D1) تحریک می شود. بیان و ترجمه ژن DRD1 باعث تولید گیرنده D1 می شود. بر این اساس این فرضیه مطرح شد که تغییرات در سطح بیان ژن DRD1 ممکن است در بروز بیماری گلوکوم نقش داشته باشد.هدف از این مطالعه بررسی بیان ژن DRD1 در بیماران مبتلا به گلوکوم بود.

    مواد و روش ها

    در این پژوهش به منظور بررسی بیان ژن DRD1 در بروز بیماری گلوکوم از جامعه آماری 40 نفره شامل 20 بیمار و 20 فرد سالم استفاده شد. پس از استخراج RNA و سنتز cDNA، از تکنیک Real-time Quantitative PCR (qRT-PCR) استفاده شد و داده های آن با نرم افزار SPSS و روش اماری من ویتنی مورد بررسی قرار گرفت.

    نتایج :

    نتایج این مطالعه نشان دهنده وجود تفاوت آماری معنی دار در میزان بیان ژن DRD1 در بیماران نسبت به گروه کنترل است.

    هدف :

    سرطان معده (GC) یک سرطان به شدت شایع در سراسر جهان و یکی از دلایل اصلی مرگ و میر ناشی از سرطان است. حساسیت افراد به سرطان معده بستگی به تغییرات ژنتیکی و اپی ژنتیکی دارد که در طول زندگی آنها رخ می دهد. C-FOS یک انکوپروتیین است که از طریق نقش انکوژنی خود در تومورزایی شرکت می کند. بسیاری از عملکردهای سلولی را تعدیل می کند و بیان نابجای آن می تواند منجر به انواع مختلفی از سرطان ها شود.

    مواد و روش ها

    روش ARMS PCR برای شناسایی انواع ژنوتیپی rs997415225 در 50 فرد سالم و 45 بیمار مبتلا به GC انجام شد. علاوه بر این، روش qRT-PCR برای تعیین بیان نسبی ژن C-FOS در 20 فرد سالم و 20 بیمار سرطانی استفاده شد.

    یافته ها

    به نظر نمی رسد که انواع ژنوتیپی rs997415225 با GC همبستگی داشته باشند (P > 0.05)، اما فراوانی آلل "A" همبستگی داشت (P = 0.048). همچنین، بین دو گروه از نظر سطح بیان ژن C-FOS تفاوت معنی داری وجود داشت (P = 0.044).

    نتیجه گیری

    مشخص شد که انواع ژنوتیپی rs997415225 تاثیری بر GC ندارند؛ با این حال، وجود آلل "A" با خطر بالقوه پیشرفت GC همراه است. علاوه بر این، افزایش بیان ژن C-FOS با پیشرفت این سرطان مرتبط بود؛ از این رو، پیشگیری از GC از طریق تنظیم پایین C-FOS ممکن است یک رویکرد امیدوارکننده باشد.

    زمینه و هدف

    بیسموت و ترکیبات مرتبط با آن مانند نانوذرات اکسید بیسموت (Bi2O3 NPs) به طور گسترده در علوم پزشکی برای دارورسانی، درمان سرطان، تصویربرداری زیستی و مبارزه با میکروارگانیسم های بیماری زا استفاده می شود. بیشتر ترکیبات شیمیایی مورد استفاده در سنتز نانوذرات برای سلامت انسان و محیط زیست سمی هستند، به همین دلیل اخیرا سنتز سبز نانوذرات مورد توجه قرار گرفته است. در روش بیوسنتز، عوامل احیا کننده شیمیایی با ترکیبات کاهنده زیستی به عنوان عامل تثبیت کننده و پوشاننده نانوذرات هستند، جایگزین می شوند.

    روش کار

    در این مطالعه، نانوذرات اکسید بیسموت (Bi2O3 NPs) با روش سنتز سبز با استفاده از عصاره سلولی سیانوباکتری Spirulina platensis تهیه شد. خواص فیزیکی و شیمیایی نانوذرات Bi2O3 با روش پراش پرتو ایکس (XRD)، تجزیه و تحلیل پرتو ایکس پراکنده انرژی (EDX)، طیف سنجی فروسرخ تبدیل فوریه (FT-IR)، میکروسکوپ الکترونی روبشی و عبوری (SEM) بررسی شد. خواص آپوپتوتیک نانوذرات بیوسنتز شده روی رده های سلولی MCF-7 و MCF-10a با بررسی اثر نانوذرات Bi2O3 بر روی بیان ژن های BAX و BCL2 و با استفاده از روش واکنش زنجیره ای پلی مراز بلادرنگ مورد آزمایش قرار گرفت.

    یافته ها: 

    اندازه، شکل و مورفولوژی نانوذرات Bi2O3 برای نفوذ به درون سلول های سرطانی و تاثیرگذاری بر فعالیت های حیاتی آنها مناسب است. در رده سلولی MCF-7، نسبت بیان BAX/BCL2 به طور قابل توجهی بالاتر از رده سلولی MCF-10a تیمار شده با نانوذرات Bi2O3 بود.

     نتیجه گیری:

    نتایج نشان دهنده تحریک آپوپتوز در رده سلولی سرطان پستان توسط نانوذرات Bi2O3 سنتز شده به روش سبز از طریق افزایش بیان ژن های تنظیم کننده آپوپتوز است.

    Objective

    Gastric cancer (GC) is the fourth most common cancer worldwide, occurring in approximately two-thirds of developing countries, and is one of the leading causes of cancer deaths. The mitogen-activated protein kinase 14 (MAPK14) plays an important role in the conversion of extracellular stimuli to a wide range of cellular responses. The 5-hydroxytryptamine receptor 2B (HTR2B) increases the level of 5-HT; and as a result, platelet-derived 5-HT promotes tumor angiogenesis, tumor growth, and metastatic potential of cancer cells.

    Materials and Methods

    Expression analysis was performed on 20 healthy cases and 20 cancerous cases for MAPK14 and 20 healthy cases and 19 cancerous cases for HTR2B using real-time quantitative PCR (qRT-PCR) assay. Polymorphisms were evaluated in 38 healthy cases and 30 cancerous cases for rs550352538, and 35 healthy cases and 35 cancerous cases for rs28763973 via tetra-primer amplification refractory mutation system PCR (T-ARMS PCR) technique.

    Results

    No significant difference was observed in MAPK14 and HTR2B gene expression levels (P>0.05). In addition, there is no statistically significant association of different genotypes in rs28763973 from MAPK14 gene and rs550352538 from HTR2B gene with gastric cancer (P>0.05).

    Conclusions

    No significant difference was noted between patient and healthy individuals in expression levels and gene polymorphisms; nevertheless, results may vary by significantly changing the gene pool or population size.

    Conservation of genetic resources of S. spicigera is important. Because the life of this valuable species has been threatened by biotic and abiotic stresses. Seed storage techniques at very low temperature (-196 °C) using liquid nitrogen is one way of maintaining germplasm with much less cost and without loss of viability on storage and it is useful when it does not lead to changes in the chemical composition of essential oil in the desired plant. In order to investigate the phytochemical stability and compare the type and amount of identified compositions, the treated seeds were transferred to the liquid nitrogen at –196 °C for 1 week as compared to the control treatment. The aerial parts of S. spicigera were collected at flowering stage and dried in the laboratory. The essential oil was isolated by hydrodistillation and analyzed by a combination of capillary GC and GC-MS. Eleven compounds were recognized in the essential oil of mentioned plant in the cryopreservation. The main constituents in the essential oil included thymol (41.1%), γ-terpinene (21.1%), p-cymene (17.2%) and carvacrol (11.9%). Results indicated that regarding the number, type and percentage of compounds existing in the essential oils, there was no difference between the control and cryopreservation treatment so by using the cryopreservation technique, the seeds of this valuable endangered species can be preserved for a long period

    Background

    Glaucoma is a term used to describe a group of diseases that progressively weaken the optic nerve and cause vision loss and eventually blindness. Glaucoma is one of the most common and controversial eye diseases. Dopamine, a neurotransmitter in the eye, regulates ciliary blood flow and aqueous humour production, which affects intraocular pressure and glaucoma. Based on previous studies on dopamine receptor ligands, it was concluded that the effect of dopamine on fluid production is stimulated by dopamine receptor 1 (D1) activation. Expression and translation of the DRD1 gene cause the production of the D1 receptor. Based on this, it was hypothesized that changes in DRD1 gene expression may play a role in the development of glaucoma. The aim of this study was to analyze DRD1 gene expression in patients with glaucoma.

    Materials and Methods

    In this study, a statistical population of 40 people including 20 patients and 20 healthy individuals was used to analyze the effect of DRD1 gene expression on patients with glaucoma. After RNA extraction and cDNA synthesis, the Real-time quantitative PCR (qRT-PCR) technique was used and its data were analyzed by SPSS software and Mann-Whitney statistical method.

    Results

    The results of this study indicate a significant difference (P = 0.015) in the expression of the DRD1 gene in patients with glaucoma compared to the control group.

    Objective

    Gastric cancer (GC) is an exceedingly prevalent cancer worldwide and one of the main reasons for death from cancer. The individuals’ susceptibility to GC depends upon several genetic and epigenetic alterations that occur over their lifetime. The C-FOS is an oncoprotein that engages in tumorigenesis through its oncogenic roles. It modulates many cellular functions, and its aberrant expression can lead to several types of cancers.

    Materials and Methods

    The Amplification Refractory Mutation System (ARMS) PCR technique was conducted to detect genotypic types of rs997415225 in 50 healthy controls and 45 patients with GC. Furthermore, the quantitative Real-Time PCR (qRT-PCR) technique was applied to determine C-FOS relative gene expression in 20 healthy controls and 20 cancer patients.

    Results

    Genotypic types of rs997415225 did not seem to be correlated with GC (P > 0.05), but allele “A” frequency was correlated (P = 0.048). Also, there was a significant difference between the two groups regarding the C-FOS gene expression level (P = 0.044).

    Conclusions

    It has been found that genotypic types of rs997415225 have no impact on GC; however, the presence of allele “A” is associated with a potential risk of GC development. In addition, the increased C-FOS gene expression was linked to the progression of this cancer; hence, preventing GC through C-FOS down-regulation might be a promising approach.

    Background and Aim

    Bismuth and its related compounds such as bismuth oxide nanoparticles (Bi2O3 NPs) are widely used in the medical field of science for drug delivery, cancer therapy, bio imaging, and fighting against pathogenic microorganisms. Most of the chemical compounds used in the synthesis of nanoparticles are toxic to human health and the environment, for this reason, recently considerable interest has been focused on the green synthesis of non-toxic and environmentally friendly nanoparticles (NPs). In the biosynthesis procedure, chemicals reducing agents are replaced by cell extract of organisms that contain bio-reductive compounds as stabilizing and capping agent for NPs.

    Methods

    In this study, bismuth oxide nanoparticles (Bi2O3 NPs) were prepared using green synthesis by cyanobacterial cell extract of Spirulina platensis. The physico-chemical properties of the Bi2O3 NPs characterized by X-ray diffraction (XRD), energy dispersive X‒ray (EDX) analysis, Fourier-transform infrared (FT-IR) spectroscopy, scanning and transmission electron microscopy (SEM). The apoptotic properties of biosynthesized nanoparticles on MCF-7 and MCF-10a cell lines were determined using the effect of the Bi2O3 NPs on the expression of BAX and BCL2 genes by real-time polymerase chain reaction.

    Results

    The size, shape, and morphology of Bi2O3 NPs are suitable for penetrating into the cancer cells and affecting their vital activities. In the MCF-7 cell line, the ratio of BAX/BCL2 expression was significantly higher than that of in MCF-10a cell line treated with the Bi2O3 NPs.

    Conclusion

    The results revealed the stimulation of apoptosis in breast cancer cell line by green synthesized Bi2O3 NPs through an increase in apoptosis regulator genes expression.
