    مقدمه:

     گونه های Phytophthora  یک تهدید جدی برای محصولهای گیاهی در دنیا هستند. بنابراین شناسایی آنها اولین قدم برای یافتن روش مدیریت بیماری است. هدف  از این مطالعه شناسایی گونه Phytophthora عامل پوسیدگی ریشه و طوقه درختان خرمالو در استان فارس بود.

    مواد و روش ها

     طوقه و ریشه درختان خرمالوی بیمار در تابستان 1399-1398، نمونه برداری شد. بافت های آلوده ریشه و طوقه در محیط CMA-PARPH  کشت داده شدند. جدایه های گونه ی Phytophthora  به روش تک اسپور خالص سازی شدند و برای شناسایی آنها، از ویژگی های ریخت شناسی و مولکولی استفاده گردید.

    یافته ها

    شش جدایه از ریشه درختان خرمالوی بیمار به دست آمدند، که بر اساس ویژگی های ریخت شناسی  Phytophthora citricola شناخته شدند. بررسی فیلوژنتیکی بر اساس ژن های بتاتوبولین و 28S rDNA،  هر شش ایزوله (Iran-Pc1 to Iran-Pc6) در کلاد 2 با مقیاس اعتبارسنجی 100 گروه بندی گردیدند و گونه P. citricola را تایید نمود.

    نتیجه گیری

    این یک گزارش جدید از پوسیدگی ریشه و طوقه خرمالو توسط Phytophthora citricola در استان فارس می باشد.

    مقدمه:

     هوانگ لونگ بینگ یا میوه سبز یکی از بیماری های مهم مرکبات در ایران است. روند گسترش بیماری در هفت استان در این تحقیق مورد بررسی قرار گرفت.

    مواد و روش ها

    باغ های مرکبات استان های سیستان و بلوچستان، هرمزگان، بوشهر، خوزستان، فارس، کرمان و مازندران طی سال های 1386 تا 1388 و 1396 تا 1398 بازدید شدند و نمونه های مشکوک به بیماری جمع آوری و پس از استخراج DNA از نمونه ها، آزمون پی سی آر با استفاده از آغازگرهای اختصاصی OI1/OI2c و A2/J5 انجام شد.

    یافته ها

    برسی نمونه ها نشان داد که، مناطق و میزبان های باکتری Candidatus Liberibacter asiaticus، عامل بیماری، در این مدت افزایش یافته است، به طوری که در پایان این دوره در نمونه های درختان پرتقال، نارنگی، گریپ فروت، نارنج، لیمولیسبون و لیموترش از استان های کرمان، هرمزگان، سیستان و بلوچستان و فارس، بیماری ردیابی شد.

    نتیجه گیری

    بررسی بیماری طی این سالها نشان داد که: مناطق آلوده و دامنه میزبانی بیماری گسترش زیادی یافته است و گسترش بیماری در مناطق کشت مرکبات در جنوب کشور با شیب ملایم بوده است. کاهش جمعیت پسیل ناقل بیماری طی این سال ها به نظر می رسد که در آهستگی روند گسترش بیماری موثر بوده است.

    مقدمه

    نماتد سیست طلایی  Globodera rostochiensis به عنوان یکی از خسارتزاترین عوامل بیماری زای سیب زمینی در دنیا محسوب می گردد. نظر به ترکیبات پوست نماتدهای سیستی و توانایی بعضی از قارچ ها در تولید آنزیم های تجزیه کننده آن، این پژوهش برای شناسایی قارچ های همراه نماتد سیست طلایی سیب زمینی اجرا شد.

    مواد و روش ها: 

    هشتاد و شش جدایه قارچی آلوده کننده نماتد سیست طلایی سیب زمینی در استان همدان در غرب ایران جداسازی و خالص سازی و بر اساس ویژگی های ریخت شناسی با کلیدهای معتبر شناسایی شدند.

    یافته ها: 

    هشت گونه قارچ متعلق به سه جنس Alternaria , Clonostachys,  Fusarium  از نماتد سیست طلایی سیب زمینی جداسازی و شناسایی شدند. بیشترین فراوانی مربوط به گونه های  Fusarium  بود.

    نتیجه گیری

    خصوصیات ریخت شناسی این هشت گونه قارچ شرح و نشان داده شده است.

    مقدمه

    قارچ های میکوریز آربوسکولی (AMF) در همزیستی با ریشه گیاهان به جذب بیشتر فسفر و افزایش رشد و نمو گیاهان کمک می کنند. هدف از این پژوهش تعیین همبستگی جمعیت و تنوع قارچهای میکوریزآربوسکولی با ویژگی های فیزیکی وشیمیایی خاک، در مزرعه های گندم  استان کهگیلویه و بویراحمد در جنوب غربی ایران، بود.

    مواد و روش ها

    سی مزرعه گندم این استان در نزدیکی زمان برداشت محصول بازدید شدند و از ریزوسفر و اندام های هوایی آن ها نمونه برداری شد. هاگهای  AMFبه روش الک سوسپانسیون ریزوسفر در آب و سانتریفیوژ کردن در محلول سوکرز جداسازی شدند. جمعیت هاگ این قارچها شمارش شدند. درصد کلنیزاسیون طول ریشه توسط این قارچها در هر نمونه محاسبه شد.  خصوصیات ریخت شناسی هاگهای جداسازی شده مطالعه شدند و اطلاعات جمع آوری شده، با توصیفهای AMF مقایسه و قارچ های حاضر در هر نمونه شناسایی شدند. بافت، pH خاک  ، فسفر خاک و موجود در گیاه تعیین شدند و ضریب همبستگی بین جمعیت و تنوع  AMFبا این خصوصیات فیزیکی و شیمیایی خاک و گیاه محاسبه شدند.

    یافته ها

      پانزده قارچ میکوریز آربوسکولی،  متلعق به 9 جنس :  Acaulospora ، Archaeospora  ، Entrophospora ، Gigaspora  ،  Rhizoglomus ، Claroideumglomus ، Funeliformis ، Septoglomus  و Scutellospora  شناسایی شدند. تنوع این قارچ ها در نمونه ها (5-)  9/2 (-2) و F. mosseae با فراوانی 90  درصد گونه غالب بود.  جمعیت این قارچ ها با pH خاک همبستگی منفی ولی با فسفر محلول خاک و فسفر گیاه همبستگی مثبت داشت. تنوع این قارچ ها نیز با  فسفر محلول خاک همبستگی مثبت داشت. بین میزان شن در خاک  و درصد کلنیزاسیون ریشه ها همبستگی مثبت وجود داشت.

    نتیجه گیری

    قارچهای میکوریز آربوسکولی در خاکهای با بافت سبک، pH کمتر همراه با مقادیر کم فسفر محلول و ماده آلی جمعیت و تنوع بیشتری دارند. همبستگی مثبت جمعیت این قارچ ها با فسفر گیاه حاکی از سودمندی آنها برای گیاه است.

    مقدمه:

     قارچهای بیمارگرگیاهی با ایجاد فساد در محصولهای گیاهی و مسمومیت های غذایی تهدیدی جدی برای امنیت غذایی جهان محسوب می شوند. یکی از روش های شناسایی قارچها مطالعه میسیلیوم آنها با میکروسکوپ الکترونی روبشی (SEM) است. برای گرفتن تصاویر میکروسکوپی با وضوح بالا و حفظ ویژگی های سطحی نمونه های میسیلیوم قارچها، آماده سازی نمونه ها یعنی روش آبگیری و خشک کردن آن ها بسیار مهم است.

    مواد و روش ها

    تاثیر دو روش آبگیری و دو روش خشک کردن میسیلیوم دو قارچ Rhizoctonia solani و Rhizopus stolonifer برای مطالعه آنها با میکروسکوپ الکترونی روبشی در این پژوهش، مقایسه شدند.

    یافته ها

    مشاهده های میکروسکوپی نشان دادند که در  R. stolonifer آبگیری با اتانول 100 درصد و خشک کردن و در R. solani در دمای اتاق و آبگیری با اتانول 50 درصد و روش انجماد نسبت به سایر روش ها تصویر صاف و با وضوح بالا تولید می کنند.

    نتیجه گیری

    آبگیری میسلیوم قارچها با اتانول 50 تا 100 درصد و خشک کردن در دمای اتاق یا منجمد کردن آن، روش های بهینه برای آمادهسازی آنها برای  مطالعه با SEM هستند و تصویر صاف و با وضوح بالا تولید میکنند و ویژگی های سطحی میسلیوم آنها حفظ شده و به خوبی قابل رویت است.

    مقدمه

    بعضی اسانس های گیاهی خاصیت ضد قارچی دارند. این پژوهش برای تعیین اثر سه اسانس گیاهی و دو قارچکش شیمیایی بر رشد پرگنه سه قارچ بیمارگر گیاهی در شرایط آزمایشگاهی انجام شد.

    مواد و روش ها

    اثر غلظت های مختلف سه اسانس آویشن (Thymus vulgaris L.)، نعناع فلفلی (Mentha piperata L.) و مرزه خوزستانی (Satureja khuzestanica Jamzad)، در مقایسه با دو قارچ‎کش مانکوزب و کاربندازیم، بر  رشد سه قارچ Alternaria solani، Botrytis cinerea  و Fusarium solani با روش رقیقسازی در محیط عصاره مخمر سوکرز مایع مورد بررسی قرار گرفتند و حداقل غلظت بازدارندگی (MIC) و حداقل غلظت قارچ‎کشی (MFC) آنها محاسبه شدند.

    یافته ها

    اسانس آویشن در بین اسانهای گیاهی بیشترین اثر بازدارندگی را بر A. solani و F. solani  و اسانس مرزه بیشترین اثر بازدارندگی را بر B. cinerea داشتند.  مانکوزب در بین قارچکشهای شیمیایی بیشترین اثر بازدارندگی را بر A. solani و کاربندازیم بیشترین اثر بازدارندگی را بر B. cinerea و F. solani داشتند.

    نتیجه‎ گیری:

     اسانس های آویشن و مرزه اثر بازدارندگی قابل ملاحظه ای بر رشد پرگنه این سه قارچ بیمارگر گیاهان دارند.

    مقدمه :

    نماتد ساقه و پیاز Ditylenchus dipsaci یک عامل بیماری زای مهم و خسارت زا در تعدادی از گیاهان زراعی و زینتی از جمله یونجه است. هدف از این پژوهش بررسی تاثیر ساپونین های خام و خالص شش اکوتیپ یونجه بر تفریخ تخم این نماتد بود تا روشی زیستی برای مدیریت آن یافت شود.  

    مواد و روش ها

    تاثیر ساپونین های خام و خالص شش اکوتیپ یونجه بر تفریخ تخم این نماتد  آزمایش در قالب طرح کاملا تصادفی به صورت فاکتوریل با دو فاکتور اکوتیپ های یونجه (شش اکوتیپ) و ساپونینهای (خام و خالص آنها) در شرایط آزمایشگاهی بررسی گردید.

    یافته ها

    تجزیه واریانس داه ها نشان داد که اثرمتقابل بین دو فاکتور اکوتیپ و ساپونین از نظر آماری معنی دار است. با غلظتهای 50 و 90 میکرولیتر ساپونین خام اکوتیپ های مختلف، به ترتیب 30 تا 42 درصد و 33 تا 59 درصد تخم های نماتد تفریخ نشدند. اکوتیپ های نیشابوری بیش ترین و پلی کراس شیراز و کریساری کم ترین تعداد تفریخ تخم را موجب شدند. غلظتهای 10 و 50 میکرولیتر ساپونین خالص به ترتیب بین 56 تا 69 درصد و 61 تا 79 درصد مجموع تخم های نماتد تفریخ نشدند. اکوتیپ های محلی میاندوآب و نیشابوری بیشترین و پلی کراس شیراز کم ترین تعداد تفریخ تخم را داشتند.

    نتیجه گیری

    ساپونینهای خالص اکوتیپهای یونجه نسبت به ساپونینهای خام تاثیر بیشتری برمیزان تفریخ تخم نماتد دارند. ساپونینهای اکوتیپ پلی کراس شیراز یونجه تاثیر بهتری نسبت به سایر اکوتیپها دارد.

    مقدمه

    نماتدها از عوامل خسارت زا در تولید خیار در جهان هستند. این پژوهش برای شناسایی نماتدهای انگل گیاهی در مزرعه ها و گلخانه های تحت کشت خیار در استان لرستان در غرب ایران انجام شد.

    مواد و روش ها

    هشتاد و پنج نمونه خاک همراه با ریشه خیار از شهرستان های مختلف استان لرستان در غرب ایران جمع آوری گردیدند. استخراج نماتدها، تثبیت و انتقال آن ها به گلسیرین و تهیه لام های میکروسکوپی دایمی و شناسایی گونه ها با استفاده از منابع علمی معتبر انجام گرفت.

    یافته ها

    بیست گونه نماتد انگل گیاهی از 12 جنس از زیر راسته Tylenchina شناسایی شدند. مشخصات ریخت شناسی Amplimerlinius globigerus و incognita Meloidogyne به عنوان زیانبارترین نماتدها شرح داده شده است.

    نتیجه گیری

    یازده گونه جدید برای فون نماتدهای خیار در ایران گزارش می شود.

    مقدمه

    نماتد غده ریشه(Meloidogyne javanica) از عوامل محدودکننده کشت محصولات گلخانه ای از جمله گوجه فرنگی محسوب می شود. این پژوهش برای بررسی تاثیر عصاره های رزماری و آنغوزه در مقایسه با نماتدکش های شیمیایی راگبی و ولوم برای یافتن روشی سازگار با محیط زیست برای مدیریت بیماری انجام شد.

    مواد و روش ها

    اثر عصاره های رزماری و آنغوزه با نماتدکش های  راگبی و ولوم بر نماتدغده ریشه گوجه فرنگی در قالب طرح کاملا تصادفی در شرایط گلخانه بررسی شد. گیاهچه های رقم موبیل در مرحله 4 تا 6 برگی با لارو سن دوم M. javanica تلقیح و اثر عصاره ها و نماتدکش ها بررسی شدند. بعد از گذشت 60 روز گیاهان از گلدان خارج و شاخصهای رشدی و شدت بیماری آن ها اندازهگیری شدند.

    یافته ها

    استفاده هم زمان عصاره های رزماری و آنغوزه بیشترین تاثیر در کاهش شاخص های بیماری زایی مانند شاخص گال، تعداد لارو و تعداد تخم و افزایش شاخص های رشدی گیاه از جمله وزن تر و خشک ریشه و وزن تر ساقه در مقایسه با شاهد آلوده به نماتد و تیمارهای نماتدکشها داشت.

    نتیجه گیری

    تاثیر خوب ترکیب عصاره های آنغوزه و رزماری در مقایسه با نماتدکشهای شیمیایی نشان از پتانسیل مناسب استفاده از آن، به عنوان روشی سازگار با محیط زیست برای مدیریت بیماری است.

    پسته یکی از مهمترین محصولات باغی در ایران بوده که تولید آن همواره تحت تاثیر عوامل محدودکننده زنده و غیر زنده قرار می گیرد. پوسیدگی آرمیلاریایی طوقه و ریشه ناشی از Armillaria mellea، یک بیماری مهم در باغ های پسته بوده که باعث مرگ و میر درختان بارور می شود. کنترل این بیماری بسیار سخت و هزینه بر می باشد. نشانه های بیماری در اندام هوایی درختان مبتلا شامل زردی، برگ ریزی زودهنگام، پژمردگی ناگهانی، سرخشکیدگی شاخه ها و کاهش رشد اندام هوایی می باشد. این نشانه ها همراه با پوسیدگی و قهوه ای شدن بافت های چوبی، تشکیل صفحات میسلیومی بادبزنی سفید تا کرمی رنگ در ریشه و طوقه و همچنین مشاهده نماریشه (رایزومورف) قارچ، شواهد کافی جهت تشخیص قطعی ابتلا به پوسیدگی آرمیلاریایی طوقه و ریشه در درختان پسته می باشند. فرآیند آلودگی با اتصال محکم ریسه و نماریشه ها روی سطح ریشه ها شروع شده و سپس بیمارگر با استفاده از روش هایی مانند فشار مکانیکی، تولید توکسین و آنزیم های تجزیه کننده دیواره سلولی به بافت های ریشه نفوذ می کند. تاخیر در تشخیص بیماری، وجود شبکه گسترده از نماریشه ها در فواصل دور از درخت و در اعماق خاک و مخفی ماندن میسلیوم ها زیر پوست و داخل چوب های مرده، باعث می شود که کنترل موفق بیماری تنها با تلفیق روش های کنترل شیمیایی، زراعی، بیولوژیکی و استفاده از پایه و ارقام مقاوم امکان پذیر باشد.

    بیماری موزاییک باکتریایی، توسط باکتری گرم مثبت Clavibacter tessellarius ایجاد می شود. از نشانه ها مهم این بیماری، موزاییک، لکه های سبز و رنگ پریده کوچک در سطح برگ می باشد که با نشانه های کمبود عناصر غذایی و برخی بیماری های ویروسی شباهت زیادی دارد و تشخیص آن را مشکل می کند. بیماری موزاییک باکتریایی، تاکنون در مزارع گندم از چندین استان در کشور گزارش شده و به نظر می رسد در سایر نواحی کشت گندم نیز وجود داشته باشد. با توجه به بذرزاد بودن این بیمارگر، تهیه بذر سالم و گواهی شده مهم ترین راهکار مدیریت این بیماری است. به منظور آشنایی بیشتر با این بیماری، جنبه های مختلف آن شامل تاریخچه، نشانه ها و روش شناسایی بیمارگر، زیست شناسی، دامنه میزبانی و مدیریت این بیماری در این مقاله شرح داده شده اند.

    جمعیت جهان به سرعت در حال افزایش است. برای تغذیه این جمعیت زیاد، باید محصولات زراعی بیشتری تولید شود. استفاده گسترده از کودها، سموم دفع آفات و سایر نهاده ها، بهره وری زمین های کشاورزی موجود و میزان غذای تولید شده را افزایش می دهد، اما این افزایش محصول باعث آسیب به محیط زیست، تخریب خاک و از بین رفتن جمعیت طبیعی میکروبی آن می شود. برای جلوگیری از این آثار زیانبار مواد شیمیایی، یک رویکرد دوستدار محیط زیست که سلامت انسان و محیط زیست را تضمین کند، به شدت مورد نیاز است. در دیدگاه نوآورانه تولید کشاورزی، برای استفاده از کودهای زیستی به جای مواد شیمیایی تقاضای فزاینده ای وجود دارد. کاربرد ریزجانداران مفید می تواند یک سازوکار جدید برای بهبود سلامت و بهره وری گیاه باشد. جمعیت های میکروبی فراریشه نقش مهمی در تبدیل و انحلال مواد مغذی خاک ایفا می کنند که برای رشد و نمو گیاه بسیار مهم است. در میان جمعیت های مختلف میکروبی فراریشه، قارچ هایی که به عنوان افزایش دهنده رشد گیاه شناخته می شوند، اخیرا مورد توجه روزافزون قرار گرفته اند. طی چندین دهه، انواع قارچ های افزایش دهنده رشد گیاه مانند گونه های متعلق به جنس های Trichoderma، Penicillium، Phoma و Fusarium مورد مطالعه قرار گرفته اند. مطالعات نشان داده است که این قارچ ها، بدون آلودگی های زیست محیطی، رشد گیاه را تنظیم می کنند و تحمل گیاه را در برابر بیمارگرهای گیاهی افزایش می دهند. در این مقاله ابتدا شرح مختصری از قارچ های افزایش دهنده رشد ارایه می شود. در ادامه ماهیت و ترکیب این قارچ ها، همچنین نحوه عملکرد آن ها، فرموله کردن و چالش های مرتبط با آن مورد توجه قرار خواهد گرفت.

    Introduction

    Phytophthora species are a serious threat to plant products worldwide. Therefore, identifying them is the first step in finding a way to treat the disease. The aim of this study was to identify Phytophthora species causing root and crown rot of persimmon trees in Fars province.

    Materials and methods

    Samples were taken from the crowns and roots of diseased persimmon trees, in the summer of 2018-2019. Infected root and crown tissues were cultured in CMA-PARPH medium. Isolates of Phytophthora species were purified by single spore method and morphological and molecular characteristics were used to identify them.

    Results

    Six isolates were obtained from the roots of diseased persimmon trees and identified as Phytophthora citricola based on their morphological characteristics. Phylogenetic studies based on beta-tubulin (βtub) and 28S rDNA genes showed that all isolates (Iran-Pc1 to Iran-Pc6) were grouped into clade 2 with a validation scale of 100 and confirmed the identification of P .citricola.

    Conclusion

    This is a new report of persimmon root and crown rot caused by Phytophthora citricola in Fars Province.

    Introduction

    Huanglongbing or Fruit Greening is one of the major citrus diseases in Iran that has spread to most of the country's citrus growing provinces. In this study, the prevalence trend of the disease in seven provinces was investigated and analyzed.

    Materials and Methods

    Citrus orchards in Sistan-Balochestan, Hormozgan, Bushehr, Khuzestan, Fars, Kerman and Mazandaran provinces were visited and samples suspected of having this disease were collected, in 2007-2009 and 2017-2019 years. After DNA extraction from the samples, the PCR test was performed using the specific primers OI1/OI2c and A2/J5.

    Results

    It was found that the percentage of infected specimens, infected areas and number of hosts of Candidatus Liberibacter asiaticus, the causative agent of this disease, has increased over this period. At the end of this period, some samples of oranges, tangerines, grapefruits, bitter oranges, Lisbon lemons and Mexican limes were obtained from the provinces: Kerman, Hormozgan, Sistan-Baluchestan, and Fars were infected with the disease.

    Conclusion

    The study of the disease during these years showed that: the infected areas and the host range of the disease have expanded a lot during this period, and the spread of the disease in citrus cultivation areas in the south of the country has been with a gentle slope. The reduction of the disease-carrying psyllium population during these years seems to have been effective in slowing down the spread of the disease.

    Introduction

    The golden cyst nematode, Globodera rostochiensis, is considered as one of the most damaging potato pathogens in the world. Considering the skin composition of cyst nematodes and the ability of some fungi to produce enzymes that decompose it, this research was conducted to identify the fungi associated with potato cyst nematode.

    Materials and Methods

    Eighty-six fungal isolates infecting potato golden cyst nematode in Hamedan province in western Iran were isolated and purified and identified based on morphological characteristics by valid keys.

    Results

    Eight species of fungi belonging to three genera Alternaria, Clonostachys, Fusarium were isolated and identified from potato golden cyst nematode. The highest frequency was related to different species of Fusarium.

    Conclusion

    The morphological characteristics of these eight fungi have been described and illustrated.

    Introduction

    Arbuscular mycorrhizal fungi (AMF) in symbiosis with plant roots help to absorb more phosphorus, and increase the growth and development of plants. The aim of this study was to determine the correlation between population and diversity of AMF, and physicochemical characteristics of soil in wheat fields of Kohgiluyeh and Boyer-Ahmad province in southwestern Iran.

    Materials and Methods

    Thirty wheat fields in this province, were visited near harvest time, and their rhizosphere and aerial organs were sampled. AMF spores were isolated by sieving the rhizosphere suspension in water and centrifuging in Sucrose solution. The spore population of these fungi was count. The percentage of root length colonization by these fungi was calculated in every sample. The morphological characteristics of the isolated spores were studied and the collected information was compared with the descriptions of AMF and the fungi present in each sample were identified. Texture, soil dispersion, soil and plant phosphorus content were determined and the correlation coefficient between AMF population and diversity with physical and chemical characteristics of soil and wheat plant were calculated.

    Results

    Fifteen arbuscular mycorrhizal fungi belonging to nine genera: Acaulospora, Archaeospora, Entrophospora, Gigaspora, Claroideumglomus, Funeliformis, Rhizoglomus, Septoglomus and Scutellospora were identified. The diversity of these fungi in the samples was (2-)2.9(-5) and F. mosseae was dominant species with 90% relative frequency. The population of these fungi had a negative correlation with soil pH, but with soil soluble phosphorus and plant phosphorus content had a positive correlation. The diversity of these fungi had a positive correlation with soil soluble phosphorus. There was a positive correlation between the amount of sand in the soil and the root length colonization.

    Conclusion

    AMF have a greater population and diversity in soils with light texture, less moisture along with low amounts of soluble phosphorus and organic matter. The positive correlation between the population of these fungi and plant phosphorus indicates their usefulness for the plant.

    Introduction

    Plant-pathogenic fungi are considered a serious threat to world food security, causing spoilage in plant products and food poisoning. One of the ways to identify fungi is to examine their mycelium using a scanning electron microscope (SEM). In order to take high-resolution microscopic images and obtain the surface properties of mushroom mycelium samples, it is very important to prepare the samples, i.e. the method of dehydrating and drying them.

    Materials and Methods

    In this research, the effect of two dehydration methods and two drying methods on the mycelia of two fungi, Rhizoctonia solani and Rhizopus stolonifer, were compared to study them with SEM.

    Results

    Microscopic observations showed that for R. stolonifer, dehydration with 100% ethanol and drying and for R. solani at room temperature and dehydration with 50% ethanol and the freezing method produce a smooth and high-resolution image compared to other methods.

    Conclusion

    Dehydration of fungal mycelium with 50% to 100% ethanol and drying at room temperature or freezing are the optimal methods to prepare them for examination with SEM and produce a smooth and high-resolution image, and the surface features of their mycelium are preserved and well-defined.

    Introduction

    Some plant essential oils have antifungal properties. This study was conducted to determine the effect of three plant essential oils and two chemical fungicides on the growth of three plant pathogenic fungi in vitro.

    Materials and methods

    The effect of different concentrations of three essential oils of thyme (Thymus vulgaris L.), peppermint (Mentha piperata L.) and khuzestani savory (Satureja khuzestanica Jamzad), compared to two chemical fungicides; mancozeb and carbendazim were investigated on the growth of three fungi Alternaria solani, Botrytis cinerea and Fusarium solani by dilution in Yeast Extract Sucrose Broth medium method and their minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) were calculated.

    Results

    Thyme essential oil had the greatest inhibitory effect on A. solani and F. solani among these essential oils, and savory essential oil had the greatest inhibitory effect on B. cinerea. Among the chemical fungicides, mancozeb had the most inhibitory effect on A. solani and carbendazim had the most inhibitory effect on B. cinerea and F. solani.

    Conclusion

    Thyme and khuzestani savory essential oils have a significant inhibitory effect on the growth of these three plants pathogenic fungi.

    Introduction

    The stem and bulb nematode, Ditylenchus dipsaci, is an important and damaging pathogen in a number of agricultural and ornamental plants, including alfalfa. The aim of this study was to investigate the effect of raw and pure saponins of six alfalfa ecotypes on the hatching of this nematode's eggs in order to find a biological method for its management.

    Materials and Methods

    The effect of raw and pure saponins of six alfalfa ecotypes on the hatching of stem nematode eggs was investigated in a completely randomized factorial design with two factors of alfalfa ecotypes (six ecotypes) and their saponins (raw and pure) in vitro.

    Results

    Analysis of variance showed that the interaction effect between two factors, ecotype and saponin, is statistically significant. At concentrations of 50 and 90 microliters of crude saponin from different ecotypes, 30-42% and 33-59% of the nematode eggs did not hatch, respectively. The Nishaburi ecotypes caused the most and the Shiraz and Khrisari polycross caused the least number of egg hatching. Concentrations of 10 and 50 microliters of pure saponin resulted in between 56 and 69% and 61 and 79% of total nematode eggs failing to hatch, respectively. The local ecotypes Miandoab and Nishaburi had the highest and Shiraz Polycross the lowest number of egg hatches.

    Conclusion

    Pure saponins of alfalfa ecotypes have a greater effect on nematode egg hatching than raw saponins. Saponins of Shiraz Polycross alfalfa ecotype have a better effect than other ecotypes.

    Introduction

    Nematodes are one of the harmful factors for cucumber production in the world. This research was conducted to identify plant parasitic nematodes in cucumber fields and greenhouses in Lorestan province in western Iran.

    Materials and Methods

    Eighty-five soil samples containing cucumber roots were collected from different cities of Lorestan province in western Iran. Extraction of nematodes, their fixation and the production of permanent microscopic slides and species identification were carried out using scientific references.

    Results

    Twenty plant-parasitic nematode species from 12 genera of the order Tylenchina have been identified. The morphological characteristics of Amplimerlinius globigerus and Meloidogyne incognita have been described as the most harmful cucumber nematodes in the province.

    Conclusion

    Eleven new species of cucumber root-associated nematode for Iran are reported here.

    Introduction

    Root-knot nematode (Meloidogyne javanica) is considered as one of the limiting factors in the cultivation of greenhouse crops, including tomatoes. This research was conducted to investigate the effect of rosemary and ferula extracts compared to chemical nematicides of rugby and volum to find an environmentally friendly method for disease management.

    Materials and Methods

    The effect of rosemary and ferula extracts with rugby and volum nematicides on tomato root-knot nematode was investigated in a completely randomized design under greenhouse conditions. Seedlings of Mobil variety were inoculated with second instar larvae of M. javanica at the 4-6 leaf stage and the effects of plants extracts and nematicides were investigated. After 60 days, the plants were removed from the pot and their growth indicators and disease severity were measured.

    Results

    The simultaneous use of rosemary and ferula extracts had the greatest effect in reducing pathogenicity indicators such as gall index, number of larvae and number of eggs and increasing plant growth indicators such as fresh and dry weight of root and fresh weight of stem compared to control infected with nematode and nematicides treatments.

    Conclusion

    The good effect of the combination of ferula and rosemary extracts compared to chemical nematicides shows the appropriate potential of its use as an environmentally friendly method for disease management.

    Pistachio is one of the most important horticultural products in Iran which its production is always affected by biotic and abiotic limiting factors. Armillaria crown and root rot caused by Armillaria mellea, is an important disease in pistachio orchards, which causes the death of fertile trees. Control of the disease is difficult, costly and rarely successful. Symptoms of the disease in shoots of affected trees includes yellowing, early defoliation, sudden wilting, dieback of branches and reduction shoot growth. These symptoms, together with the decay and browning of woody tissues and the formation of white to creamy mycelial sheets in the roots and crown, as well as the observation of rhizomorphs of the fungus, are sufficient evidence for definitive diagnosis of Armillaria crown and root rot in pistachio trees. The infection process begins with the growth of hyphae or rhizomorphs and their firm attachment to the surface of the roots. The pathogen uses a combination of methods such as mechanical pressure, toxin production, and cell wall-degrading enzymes to penetrate the root tissues. Successful control of the disease is possible only by combining chemical, cultural, biological control methods and use of resistant rootstocks and cultivars due to delay in diagnosis of the disease before the spread of the pathogen in plant and soil and the appearance of symptoms, the presence of an extensive network of rhizomorphs at a distance from the tree and deep in the soil and formation of the mycelium beneath the plant bark or inside dead wood.

    Bacterial mosaic of wheat is caused by the gram positive bacterium Clavibacter tessellarius. The symptoms of the disease are uniformly distributed small chlorotic spots with mosaic pattern on the entire leaf surface resemble those symptoms of nutrient deficiencies and some viral diseases, and it may be difficult to determine. Due to the seed-borne nature of the disease, use of healthy and certified pathogen free seeds is the most important strategy for managing this disease. The wheat bacterial mosaic has been reported in wheat fields in several provinces of Iran and it seems to be widespread in different wheat growing areas. In order to better understand the disease, its various aspects including the symptoms, pathogen biology and its host ranges, and management of the disease are reviewed in this article.

    The world's population is growing rapidly. To feed this large population, more crops must be produced. The broad use of fertilizers, pesticides and other inputs increases the productivity of existing agricultural lands and the amount of food production, but this increase in yield causes damage to the environment, soil degradation and the destruction of its natural microbial population. To prevent these harmful effects of the chemicals, an eco-friendly approach is needed to ensure human health and the environment. In the innovative view of agricultural production, there is a growing demand for the use of biofertilizers instead of agrochemicals. The use of beneficial microorganisms can be a new strategy to improve plant health and productivity. Natural microbial populations play an important role in the solubilizing and mineralizing of soil nutrients, which is very important for plant growth and development. Among the various microbial populations, fungi known as plants growth promoting fungi (PGPF) have recently received increasing attention. For decades, plant growth promoting fungi such as Trichoderma, Penicillium, Phoma, Aspergillus and Fusarium have been studied. Studies have shown that these fungi, regulate plant growth without environmental pollution and increase plant tolerance to plant pathogens. In this review a brief description of plant growth promoting fungi is given first. In the following, the nature and composition of these fungi as well as the mode of actions, formulation and related challenges will be concidered.
