بررسی ارتباط برخی از پارامترهای بیوشیمیایی سرم با وقوع کیست تخمدانی در گاوداری های شیری شهرستان اراککیست های تخمدانی از جمله مهم ترین عوارضی هستند که منجر به اختلالات تولیدمثلی و خسارات اقتصادی در گله های گاو شیری می شوند. مطالعه حاضر به منظور بررسی پارامترهای بیوشیمیایی سرم خون شامل گلوکز، فسفر، پروتیین تام، نیتروژن اوره ای خون (BUN)، تری گلیسیرید و همچنین برخی از خصوصیات بالینی در مبتلا به کیست تخمدانی و گاوهای سالم انجام گردید. در این مطالعه، تعداد 80 راس گاو ماده در دو گروه مساوی شامل40 گاو مبتلا به کیست تخمدانی و 40 گاو سالم تقسیم بندی شده و سپس نمونه های سرمی و اطلاعات بالینی آن ها جمع آوری گردید. مطالعه حاضر نشان داد که اغلب کیست های تخمدانی از نوع لوتیال بوده و آنستروس، شایع ترین علامت بالینی مشاهده شده در گاوهای مبتلا به کیست های تخمدانی می باشد. همچنین، ارزیابی پارامترهای بیوشیمیایی سرم نشان داد که میان گاوهای مبتلا به کیست های تخمدانی و گاوهای سالم از نظر مقادیر BUN، فسفر و پروتیین تام سرم تفاوت معنی داری وجود ندارد، در حالی که این تفاوت در خصوص مقادیر گلوکز و تری گلیسیرید معنی دار بود. بر اساس داده های این بررسی می توان نتیجه گیری نمود که بالانس منفی انرژی می تواند نقش مهمی در بروز کیست های تخمدانی در منطقه ی مورد مطالعه داشته باشد.کلید واژگان: کیست تخمدانی, گلوکز, گاو شیری, تریگلیسیرید, کاهش باروریOvarian cyst is one of the most important diseases that leads to reproductive disorders and many economic losses in dairy cows. The aim of the present study was to evaluate the blood serum biochemical parameters including glucose, phosphorus, total protein, blood urea nitrogen and triglyceride, and also some clinical features in cows with ovarian cysts and compare them with healthy cows. In this study 80 dairy cows were divided to 2 groups of cows with ovarian cysts and healthy cows that contain 40 cows, then blood samples and their clinical information were collected. The results of the present study showed that the most type of the ovarian cysts are luteal, and anestrus is the most common clinical symptom observed in cows with ovarian cysts. Furthermore, evaluation of serum biochemical parameters showed that there were no significant differences in BUN, phosphorus and total serum protein levels between cystic and healthy cows, while glucose and triglycerides levels were significantly different. It can be concluded that low glucose and high triglyceride levels play an important role in the development of ovarian cysts in the study area.Keywords: dairy cow, Subfertility, glucose, Ovarian Cyst, Triglyceride

BackgroundThe objective of the study was to investigate the effects of in vitro zinc sulphate additive to semen extender on sperm parameters (progressive motility, viability, membrane integrity and DNA stability) after cryopreservation.Materials And MethodsIn this Prospective longitudinal laboratory study, semen samples of 5 buffalo bulls of 3-5 years old were collected at 5 different occasions from Iran, Urmia during summer and autumn 2011, 25 samples were used in each treatment. Sperm progressive motility, viability and abnormal morphology were measured before and at 0.5 (T0), 1(T1) and 2(T2) hours after diluting semen(1:10 v/v) in Tris-citric acid based extender (without egg yolk and glycerol) at 37˚C containing none (control group), 0.072, 0.144, 0.288, 0.576 and 1.152 mg/L zinc sulphate to investigate dose and time effects. Next, a Tris-citric acid-egg yolk-glycerol extender (20% egg yolk and 7% glycerol) containing the same amount of zinc sulphate was prepared, diluted semen (1:10 v/v) was cooled and kept into a refrigerated chamber (4˚C) for 4 hours to equilibrate. Sperm progressive motility, viability, abnormal morphology, membrane integrity and DNA damage were estimated.The equilibrated semen was loaded in 0.5 ml French straws and frozen in liquid nitrogen. Later, the frozen semen was thawed and the same parameters as well as total antioxidant capacity (TAC) of the frozen-thawed semen were determined.ResultsThe results showed that zinc sulphate additive at the rate of 0.288 mg/L gave a higher protection of sperm progressive motility (53.7 ± 1.8% vs. 40.5 ± 1.7%), viability (70.8 ± 1.8% vs. 60.1 ± 1.5%), membrane integrity (67.3 ± 1.6% vs. 56.6 ± 1.7%), DNA stability (10.1 ± 0.47% vs. 11.8 ± 0.33% damaged DNA) through the process of dilution,equilibration and freeze-thawing and caused a higher TAC level (81 ± 3.3% vs. 63 ± 3.2 μmol/L) after freez-thawing compared to the control group. Adding 0.576 and 1.152 mg/L zinc sulphate, however, was deleterious to the sperm and significantly reduced the studied sperm parameters.ConclusionAdding 0.288 mg/L zinc sulphate to the extender, compared to the control group, gives a better sperm preservation upon freezing processes which in turn, may results in higher semen fertility. But, addition of higher zinc sulphate concentrations (0.576 and 1.152 mg/L) are detrimental to buffalo spermatozoa.Keywords: Semen, Zinc, Buffalo

To investigate the IgG content and its variations in uterine fluid (UF) during the estrous cycle of the cow and to compare them with those of the blood serum (S), six pairs of serum and UF samples for each phase of the cycle selected out of 240 bovine genital tracts and blood samples were collected from Urmia abattoir. The UF samples were collected by gentle scraping of the endometrium using a curette after uterine incision and their IgG content and those of the serum were measured by single radial immuno-diffusion (SRID) assay. Serum IgG values (Mean ± SEM) were generally higher than the UF values throughout the cycle except for di-estrus (S: 38.50 ± 0.90, UF: 51.60 ± 2.10 mg mL-1), in which the highest values were observed in UF samples. In met-estrus the difference was not significant (S: 34.80 ± 1.80mg mL-1, UF: 30.80 ± 5.20 mg mL-1), however, in estrus the mean UF IgG value (12.50 ± 1.10 mg mL-1) was lower than that of the serum (31.30 ± 1.20 mg mL-1). In pro-estrus, the lowest values (S: 27.80 ± 1.30 mg mL-1, UF: 9.10 ± 1.50 mg mL-1) were obtained. The results showed a lower IgG values in the bovine UF than those of the serum in the follicular phase of the cycle, while in di-estrus the UF IgG content was the highest, suggesting some IgG production in the uterus at this phase.Keywords: Cow, Estrous cycle, Immunoglobulin G, Uterine fluid

The aim of the present study was to investigate the effect of in vitro supplementation of selenium on fresh and frozen spermatozoa quality of buffalo (Bubalus bubalis) bulls. Five healthy buffalo bulls (5 ejaculates from each bull) were used. Each ejaculate was diluted at 37 ˚C with tris-based extender containing 0 (control)، 0. 5، 1، 2، 4 and 8 μg mL-1 sodium selenite and the sperm motility and viability were evaluated at 0 (T0) (immediately after dilution)، 60 (T1) and 120 (T2) min after diluting semen. In the second step، semen samples were diluted with tris-egg yolk-glycerol extender containing the same amounts of sodium selenite، cooled to 4 ˚C، equilibrated and semen parameters (motility، viability، membrane integrity and DNA damage) were estimated. Then، the semen was packed in 0. 5 mL French straws and frozen in liquid nitrogen. Later، the semen was thawed and analyzed for the same parameters، as well as total antioxidant capacity. Results showed that addition of 1 and 2 μgmL-1 selenium to the semen extender significantly increased the sperm motility of fresh and equilibrated semen compared to the control without affecting other parameters. However، in frozen-thawed semen، extenders containing 1 and 2 μg mL-1 selenium significantly improved sperm motility، viability، membrane integrity and semen total antioxidant capacity and also resulted in lower DNA damaged sperms. In this study selenium supplementation of semen extender of 4 and 8 μg mL-1 had deleterious effects on sperm parameters as early as the samples were prepared for freezingKeywords: Buffalo, Semen, Selenium supplementation
