The present systematic review of animal studies on long-term fructose intake in rodents revealed a significant decrease in the activities of antioxidant enzymes due to a fructose-rich diet. The reduced activity of these enzymes led to an increase in oxidative stress, which can cause liver damage in rodents. Of eight studies analyzed, 5 (62.5%) and 1 (12.5%) used male and female rats, respectively, while 2 studies (25%) used female mice. Moreover, half of the studies used high-fructose corn syrup, but the other half employed fructose in the diet. Hence, it is essential to monitor dietary habits to ensure public health and nutrition research outcomes.

BackgroundTrastuzumab is a humanized monoclonal antibody that targets site-specifically human epidermal growth factor-2 receptor (HER2) cell surface antigen overexpressed in approximately 20% of human breast carcinomas. Despite its positive therapeutic outcomes, a large proportion of individuals are unresponsive to the treatment with trastuzumab or develop resistance to it.ObjectiveTo evaluate a chemically synthesized trastuzumab-based antibody-drug conjugate (ADC) to improve the trastuzumab therapeutic index.MethodsThe current study explored the physiochemical characteristics of the trastuzumab conjugated to a cytotoxic chemotherapy agent DM1 via Succinimidyl 4-(N-maleimidomethyl) cyclohexane-1-carboxylate (SMCC) linker, created in our earlier study, using SDS-PAGE, UV/VIS, and RP-HPLC analyses. The antitumor effects of the ADCs were analyzed using MDA-MB-231 (HER2-negative) and SK-BR-3 (HER2-positive) cell lines utilizing in vitro cytotoxicity, viability, and binding assays. Three different formats of a HER2-targeting agent: trastuzumab, synthesized trastuzumab-MCC-DM1, and commercially available drug T-DM1 (Kadcyla®) were compared.ResultsUV-VIS spectroscopic analysis showed that the trastuzumab-MCC-DM1 conjugates, on average, entailed 2.9 DM1 payloads per trastuzumab. A free drug level of 2.5% was determined by RP-HPLC. The conjugate appeared as two bands on a reducing SDS-PAGE gel. MTT viability assay showed that conjugating trastuzumab with DM1 significantly improved the antiproliferative effects of this antibody in vitro. Importantly, the evaluations using LDH release and cell apoptosis assays confirmed that trastuzumab maintains its ability to induce cell death response while conjugating with the DM1. The binding efficiency of trastuzumab-MCC-DM1 was comparable to that of the naked trastuzumab.ConclusionTrastuzumab-MCC-DM1 was found effective against HER2+ tumors. شربت ذرت با فروکتوز بالا-55 (55-HFCS)، شیرین کننده متداولی در صنایع غذایی است. اخیرا اثرات سوء مصرف خوراکی بلند مدت کنترل نشده 55-HFCS در مقایسه با سوکروز بر ناکارآمدی تخمدان موش صحرایی ماده را منتشر کرده ایم. هدف این مطالعه مقایسه اثرات مصرف کنترل شده کوتاه مدت 55-HFCS با سوکروز و تاثیرات احتمالی آن بر بافت رحم و تخمدان موش های صحرایی ماده در مقایسه با مصرف بلند مدت آن بود.
روش بررسیطبق گایدلاین OECD (شماره 407)، 15سر موش صحرایی ماده به 3 گروه تقسیم شدند. گروه کنترل دریافت کننده آب مقطر، گروه دوم دریافت کننده سوکروز 75 % و گروه سوم دریافت کنندهHFCS-55 به میزان g 100/ml 1 وزن بدن به شکل گاواژ (28 روز) بودند. الگوی دریافت انرژی، وزن، تغییرات رفتاری و شاخص های بیوشیمیایی و هیستوپاتولوژی در روز 29 مورد ارزیابی قرار گرفت.
یافته ها:
هیچ گزارش مرگ و میر، تغییرات در الگوی انرژی، وزن و رفتار دیده نشد. از بین پروفایل لیپیدی و فاکتورهای بیوشیمیایی، صرفا سطح سرمی اسید اوریک و HDL در گروه های HFCS-55 و سوکروز نسبت به کنترل کاهش معنی داری یافت. درگروه HFCS-55 آپوپتوز سلولی در بافت رحم و احتقان بافت تخمدان ثبت شد که مشابه الگوی مطالعه 90روزه بود.
نتیجه گیری:
نتایج این مطالعه در تطبیق با مطالعه 90 روزه، دلالت بر آسیب پذیری بافت رحم و تخمدان نسبت به فروکتوز در مقایسه با سوکروز دارد. خطر آسیب احتمالیHFCS-55 بر رحم و تخمدان زنان نیاز به مطالعات تکمیلی دارد.

Recently, we have investigated and published the effects of uncontrolled long-term oral ingestion of 55-HFCS compared to sucrose on primary ovarian insufficiency in female rats. This study aimed to compare the effects of short-term controlled HFCS-55 administration with sucrose and its possible effects on the uterine and ovarian tissue of female rats compared to the long-term model.
Materials and methodsAccording to OECD guideline (No. 407), 15 female rats were divided into three groups. The control group received distilled water; the second group received sucrose 75%; the third group received HFCS-55; 1ml/100 g body weight, for 28 days by gavage. The pattern of energy intake, weight, behavioral changes, and indicators of biochemical and histopathology were evaluated on day 29.
ResultsThere were no reports of death, or changes in energy intake, weight, and behavior. Out of lipid profile and biochemical factors, serum levels of uric acid and HDL in the HFCS-55 and sucrose groups were significantly reduced compared to the control. In the HFCS-55 group, cell apoptosis was recorded in the uterine tissue (similar to the 90-day study), and ovarian tissue congestion was reported like in the 90-day study.
ConclusionResults of our study compared to the 90-day study, indicate the vulnerability of the uterine and ovarian tissues caused by fructose. The possible damage risk of HFCS-55 on the uterus and ovary of women needs further epidemiological studies.
